目的

探索酒精对慢性前列腺炎/慢性盆腔疼痛综合征(CP/CPPS)中T淋巴细胞变化的影响。

方法

皮下多点注射自身抗原法建立实验性自身免疫性前列腺炎(EAP)模型模拟CP/CPPS。酒精处理组同时给予液体酒精饲料饮食,安慰剂组予安慰剂饮食。42 d后,通过HE染色评估前列腺组织的炎症表现,疼痛学评分评估骨盆疼痛的严重程度。通过流式细胞学检测小鼠脾脏的Th1、Th2、Th17、Treg细胞比例的变化情况。

结果

HE染色显示,与EAP+安慰剂组相比,EAP+酒精组小鼠的前列腺组织呈现出更加严重的弥漫性炎症,主要表现为严重的嗜中性粒细胞和单核细胞的浸润。同时,流式细胞学检测可以明显地观察到EAP+酒精组小鼠的Th1细胞和Th17细胞比例明显升高,Th2细胞和Treg细胞比例明显下降。

结论

酒精能够加重EAP小鼠的前列腺炎症,且引起淋巴细胞Th1/Th2及Th17/Treg比例失衡,这可能在酒精加重EAP前列腺炎症中发挥重要作用,为CP/CPPS的诊断治疗提供一种新的思路。

Objective

To explore the changes of alcohol on T lymphocytes in chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS).

Methods

The experimental autoimmune prostatitis (EAP) model was established by subcutaneous multi-point injection of autoantigen to simulate CP/CPPS. The alcohol-treated group was given a liquid alcohol diet and the placebo group was given a placebo diet. After 42 days, the inflammatory manifestations of prostate tissue were assessed by HE staining, and the severity of pelvic pain was assessed by pain score. The changes of Th1, Th2, Th17 and Treg cells in mouse spleen were detected by flow cytometry.

Results

HE staining showed that compared with the EAP+placebo group, the prostate tissue of the EAP+alcohol group showed more severe diffuse inflammation, mainly manifested by severe neutrophil and monocyte infiltration. At the same time, flow cytometry can clearly observe that the proportion of Th1 cells and Th17 cells in the EAP+alcohol group mice was significantly increased, and the proportion of Th2 cells and Treg cells was significantly decreased.

Conclusions

This study shows that alcohol can aggravate prostate inflammation in EAP mice, and alcohol induces the imbalance of Th1/Th2 ratio and Th17/Treg ratio of lymphocytes, which may play an important role in alcohol aggravation of EAP prostate inflammation, and provide a new way of thinking for the diognosis and treatment of CP/CPPS.
