Comparison of the efficacy of composite scaffolds with different proportions of polylactic acid and silk fibroin in urethral repair in rabbits

Abstract:

Objective

To compare the effect of 3D printing composite scaffolds with different proportions of polylactic acid (PLA) and silk fibroin (SF) in urethral repair in rabbits.

Methods

The composite scaffolds were built with different ratios of PLA and SF (25∶75, 50∶50, 75∶25), and the Urethral epithelial cells that from stem cells were co-cultured on the composite scaffolds. 24 male rabbits with artificial hypospadias were randomly divided into 3 groups, and treated by urethroplasty with different composite scaffolds. Urethral grafts were collected for histological analysis 6 weeks after urethroplasty, and the incidence of urethral stricture was calculated.

Results

As the ratios of SF in the composite increased, the number of cells that adhered to the composite and propagated on the composite increased in vitro. Six weeks after urethroplasty, the incidence of urethral stricture in the group that proportion of PLA and SF 25∶75 was lower than that in the other two groups (P<0.05).

Conclusion

The composite scaffolds, which proportion of PLA and SF was 25∶75, were more helpful for urethral defect repair.
