A preliminary study of the diagnostic value of urine extracellular vesicle circRNA classifier for prostate cancer

Abstract:

Objective

To examine the efficacy of the urine extracellular vesicle circRNA classifier (Ccirc) in patients with PSA>2 ng/ml to evaluate its potential application in prostate cancer diagnosis.

Methods

Twenty-three high-grade prostate cancer cases and 17 low-grade prostate cancer/prostatic hyperplasia cases were screened to measure extracellular vesicle circRNA expression in their urine using third-generation droplet digital PCR technology. The pathological results of prostate biopsy (biopsy) were used as judging criteria, and the effect of urine extracellular vesicle circRNA classifier (Ccirc) and PSA in the diagnosis of high-grade prostate cancer was evaluated using the receiver working characteristics (ROC) curve.

Results

The expression of Ccirc in the high-grade prostate cancer group and the low-grade prostate cancer/prostatic hyperplasia group were (7.78±1.04) copies/ml and (6.63±1.18) copies/ml, respectively, and the Ccirc expression in the high-grade prostate cancer group was higher than that in the low-grade prostate cancer/prostatic hyperplasia group (P=0.003). The area under the ROC curve for the diagnosis of high-grade prostate cancer by urine extracellular vesicle circRNA classifier and PSA was (95%CI 0.596-0.878) and 0.586(95%CI 0.419-0.739), respectively.

Conclusion

The urine extracellular vesicle circRNA classifier (Ccirc) has good efficacy in diagnosing high-grade prostate cancer patients with PSA>2 ng/ml.
